Description

Description: This assay employs a two-site sandwich ELISA to quantitate FZD6 in samples. An antibody specific for FZD6 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any FZD6 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for FZD6 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of FZD6 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Overview: The FZD6 represents a member of the 'frizzled' gene family, which encode 7-transmembrane domain proteins that are receptors for Wnt signaling proteins. The protein encoded by this family member contains a signal peptide, a cysteine-rich domain in the N-terminal extracellular region, and seven transmembrane domains, but unlike other family members, this protein does not contain a C-terminal PDZ domain-binding motif.
This protein functions as a negative regulator of the canonical Wnt/beta-catenin signaling cascade, thereby inhibiting the processes that trigger oncogenic transformation, cell proliferation, and inhibition of apoptosis. Alternative splicing results in multiple transcript variants
